Clinical application value of LncRNA ANRIL expression in plasma of patients with acute ischemic stroke

LncRNA ANRIL在急性缺血性脑卒中患者血浆中的表达及临床应用

Abstract: Objective To investigate the Long noncoding RNA antisense noncoding RNA in the INK4 locus in plasma of patients with acute ischemic brain death(AIS).Correlation between LncRNA ANRIL expression level and inflammatory factors IL-10 and TNF-α and its clinical application value in nerve function injury.Methods Plasma samples and general clinical data were collected from 180 patients diagnosed with acute ischemic stroke(AIS group) and 130 healthy controls(NC group) hospitalized in the Department of Neurosurgeryof our Hospital from January 2019 to January 2021.The expression levels of LncRNA ANRIL in plasma samples were detected by real-time fluorescence quantitative PCR(QRT-PCR),and the expression levels of IL-10 and TNF-α in plasma samples were detected by ELISA.Patients with acute ischemic stroke were scored according to the National Institutes of Health Stroke Scale(NIHSS).Results The expression of LncRNA ANRIL in AIS group was significantly lower than that in control group(T=8.499,P<0.001).Spearman correlation analysis showed that,The expression of LncRNA ANRIL in AIS patients was negatively correlated with NIHSS score(r=-0.7013,P<0.001),and positively correlated with IL-10 expression(R =0.3215,P<0.05).TNF-α expression was negatively correlated with TNF-α expression(r=-0.3278,P<0.05).Conclusion The decrease of plasma LncRNA ANRIL expression level in AIS patients is correlated with inflammatory factors and stroke severity,suggesting that LncRNA ANRIL may be involved in regulating the progression of AIS by regulating the expression level of inflammatory cytokines.

摘要: 目的 探讨急性缺血性脑卒患者(AIS)血浆中长链非编码RNA INK4位点反义非编码RNA(Long noncoding RNA antisense noncoding RNA in the INK4 locus,LncRNA ANRIL) 表达水平与炎症因子IL-10、TNF-α的相关性及在神经功能损伤中的临床应用价值。方法 收集2019 年1月至 2021 年1月,在我院神经外科住院的180例确诊为急性缺血性脑卒中患者(AIS 组)及130例健康对照组(NC 组)血浆样本及临床一般相关资料。分别采用实时荧光定量PCR(qRT-PCR)检测血浆样本中的LncRNA ANRIL表达量,及ELISA方法检测血浆样本中的IL-10、TNF-α的表达水平,并按美国国立卫生研究院卒中表(NIHSS)标准对急性缺血性脑卒中患者病危程度进行评分。结果 AIS组患者血浆中LncRNA ANRIL表达量较将抗对照组明显降低(t=8.499,P<0.001),Spearman 相关性分析显示,LncRNA ANRIL在AIS患者血浆中的表达量与NIHSS评分呈负相关(r=-0.7013,P<0.001),并且与IL-10表达量呈正相关性(r=0.3215,P<0.05),相反,与TNF-α表达量呈负相关性(r=- 0.3278,P<0.05)。结论 AIS 患者血浆LncRNA ANRIL 表达水平的降低与炎症因子及脑卒中严重程度相关,提示 LncRNA ANRIL 可能通过调节炎症细胞因子的表达水平参与调控 AIS 的病程进展。
