癫痫儿童miR-15a-5p、miR-125a表达意义及诊断价值分析

Expression significance and diagnostic value of miR-15a-5p and miR-125a in children with epilepsy

摘要: 目的 探讨癫痫患儿微小核糖核酸-15a-5p(miR-15a-5p)、miR-125a的表达水平,分析二者对儿童癫痫的诊断价值。方法 纳入我院2017年9月~2021年4月收治的癫痫患儿82例作为癫痫组,取同期于我院体检的健康儿童65例为对照组。检测两组血清miR-15a-5p、miR-125a表达以及白细胞介素-1β(IL-1β)、IL-2、肿瘤坏死因子-α(TNF-α)水平,分析癫痫患儿miR-15a-5p、miR-125a与IL-1β、IL-2、TNF-α的相关性及miR-15a-5p与miR-125a的相关性。分析癫痫儿童miR-15a-5p、miR-125a表达与临床特征的关系,绘制受试者工作特征曲线(ROC)评估二者诊断儿童癫痫的曲线下面积(AUC)。结果 癫痫组血清miR-15a-5p、miR-125a表达低于对照组,IL-1β、IL-2、TNF-α水平高于对照组(P<0.05)。Pearson线性相关检验提示癫痫患儿血清miR-15a-5p、miR-125a与IL-1β、IL-2、TNF-α水平呈负相关,miR-15a-5p与miR-125a呈正相关(P<0.05)。miR-15a-5p、miR-125a低表达组频繁发作、症状持续时间>10 min、全面性发作占比高于miR-15a-5p、miR-125a高表达组(P<0.05)。血清miR-15a-5p、miR-125a单独与联合诊断儿童癫痫的AUC分别为0.773、0.756、0.862。结论 癫痫患儿血清miR-15a-5p、miR-125a表达降低,二者表达与炎症因子密切相关,对儿童癫痫有一定诊断价值。

Abstract: Objective To explore the expression levels of microRNA-15a-5p(miR-15a-5p) and miR-125a in children with epilepsy,and analyze their diagnostic value in children with epilepsy.Methods A total of 82 children with epilepsy admitted to our hospital from September 2017 to April 2021 were included as the epilepsy group,and 65 healthy children who received physical examination in our hospital during the same period were selected as the control group.To detect the expression of serum miR-15a-5p,miR-125a and the levels of interleukin-1β(IL-1β),IL-2,and tumor necrosis factor-α(TNF-α) in the two groups.To analyze the correlation between miR-15a-5p,miR-125a and IL-1β,IL-2,TNF-α and the correlation between miR-15a-5p and miR-125a in children with epilepsy.To analyze the relationship between the expression of miR-15a-5p,miR-125a and clinical features in children with epilepsy.A receiver operating characteristic curve(ROC) was drawn to evaluate the area under the curve(AUC) of the two in diagnosing childhood epilepsy.Results The expressions of serum miR-15a-5p and miR-125a in the epilepsy group were lower than those in the control group,and the levels of IL-1β,IL-2,and TNF-α were higher than those in the control group(P<0.05).Pearson linear correlation test showed that serum miR-15a-5p and miR-125a in children with epilepsy were negatively correlated with IL-1β,IL-2,TNF-α levels,and miR-15a-5p was positively correlated with miR-125a(P<0.05).The miR-15a-5p and miR-125a low expression group had frequent attacks,the duration of symptoms was more than 10 minutes,and the proportion of generalized attacks was higher than that in the miR-15a-5p and miR-125a high expression group(P<0.05).The AUC of serum miR-15a-5p and miR-125a alone and combined with the diagnosis of childhood epilepsy were 0.773,0.756,0.862,respectively.Conclusion The expression of serum miR-15a-5p and miR-125a in children with epilepsy is decreased,and their expression is closely related to inflammatory factors,which has a certain diagnostic value for children with epilepsy.
